چکیده

The importance of dopamine in the motor functions of the striatum is evident in Parkinson’s disease (PD). The striatum controls motor activity by processing the flow of information arising from the cerebral cortex and projecting via direct and indirect pathways to the output nuclei of the basal ganglia. The degenerative loss of dopamine is a hallmark of this disease and leads to severe motor impairments that are relieved by dopamine agonists. However, dopamine plays a role not only in the execution of complex movement, but also in higher-order cognitive processes, including motor planning and sequencing, motor learning, and motivational drive and affect. Of the biogenic amine neurotransmitters, dopamine has been the best studied in the central nervous system (CNS). The actions of dopamine are segregated in different neural circuits. For example, dopamine in the nigrostriatal pathway is involved in the generation and execution of voluntary movement. In this function, dopamine is a prime modulator of

P139: Role of Dopamine Receptor D3 in Depression and Anxiety

Dopamine (DA) is one of the main catecholamines in the brain and is crucial for movement coordination, endocrine function, reward, mood, memory and emotions. The dopaminergic system is the primary therapeutic target in the treatment of Parkinson’s disease (PD), drug addiction and schizophrenia. Notwithstanding, dysfunction of central dopaminergic neurotransmission has also been associated to de...

Cinnamaldehyde attenuates dopaminergic neuronal loss in substantia nigra and induces midbrain catalase activity in a mouse model of Parkinson’s disease

Background and Objective: Parkinson's disease (PD) is the second most common neurodegenerative disease after Alzheimer's disease that affects 3% of the population. PD involves a progressive degeneration of dopaminergic neurons in the substantia nigra pars compacta (SNc) and subsequent loss of dopamine. Dopamine depletion leads to movement dysfunction and is accompanied with tremor, rigid muscle...

Dopamine-Synthesizing Neurons: An Overview of Their Development and Application for Cell Therapy

Cell-gene therapy is a dynamic constituent of novel medical biotechnology. Neurodegenerative disordersin which damage to or demise of specific brain cell types plays central role, are clear examples of diseasecandidate for cell replacement therapy. Dopaminergic (DAergic) neurons biosynthesize dopamine, a vitalneurotransmitter in the central nervous system. Due to the involveme...

Multiple dopamine receptors: relevance for neurodegenerative disorders.

In several neurodegenerative disorders such as Parkinson’s disease and Huntington’s disease, neuronal systems employing the neurotransmitter dopamine have been implicated in either the pathology or the drug treatments of the disease (for review see [ 11). It is therefore of some importance to understand the mechanism of action of dopamine in controlling brain function, both in the normal state ...

Signaling with Increased Nigrostriatal Damage: Relevance to Parkinson’s Disease

Nicotinic receptors (nAChRs) are important modulators of dopaminergic transmission in striatum, a region critical to Parkinson’s disease. The nAChRs mainly involved are the 6 2* and 4 2* subtypes. Lesion studies show that the 6 2* receptor is decreased to a much greater extent with nigrostriatal damage than the 4 2* subtype raising the question whether this latter nAChR population is more impor...
